The immunoglobulin superfamily (IgSF) cell adhesion molecules are calcium-independent transmembrane glycoproteins, including: neural cell adhesion molecules (NCAMs), intercellular cell adhesion molecules (ICAMs), vascular cell adhesion molecule (VCAM), platelet-endothelial cell adhesion molecule (PECAM-1), endothelial cell-sel ective adhesion molecule (ESAM), junctional adhesion molecule (JAMs), nectins, and other cell adhesion molecules. Each IgSF CAM has an extracellular domain, which contains several Ig-like intra-chain disulfide-bonded loops with conserved cysteine residues, a transmembrane domain, and an intracellular domain that interacts with the cytoskeleton. They are either homophilic or heterophilic and bind integrins or different IgSF CAMs.The best-studied members of the IgSF cell adhesion molecules are the NCAMs which are expressed predominantly in nervous tissue and the ICAMs. NCAMs play important roles in an array of development progresses, including cell migration, proliferation, and differentiation. Recent research findings suggest that NCAM signal in part through integrins leading to cytoskeletal rearrangements locally in the growth cone or cell leading edge, and to MAP kinase. ICAM-1 and VCAM-1 have an essential role in monocyte recruitment. It has also been suggested that cell adhesion molecules may have a role in the pathogenesis of atherosclerotic vascular disease in diabetes.
